The combination of fresh flash-frozen tissue and laser microdissectionmadeitpossibletoisolateRNAfromtheepithelialcelllayer,only.Inthesmallintestine,Lgr5labelscellsare inthe+4position,whileinthecolon,Lgr5positivecellsarelocalizedtothecryptbottoms.Epithelialcryptcellsofnormal smallintestineexpressedneitherEP-receptormRNAnorCOX1/2.However,cryptcellsintissuefrompatientswithuntreated celiacdiseaseexpressedEP2/4receptorandCOX1mRNA.Inthecolon,thesituationwasdifferent.Epithelialcryptcellsfrom normalcolonwerefoundtoexpressEP2/4receptorandCOX1/2transcripts.Thus,therearedistinctdifferencesbetween normalhumansmallintestineandcolonwithregardtoexpressionofEP2/4receptorsandCOX1/2.Innormalcolontissue, PGE2-mediated signaling through EP-receptors 2/4 could be involved in regulation of growth and differentiation of the epithelium,whilethelackofEP-receptorexpressioninthesmallintestinaltissueexcludethepossibilityofadirecteffectof PGE2onthecryptepithelialcells.
