We show here that the budding yeast Ctf19 kinetochore subcomplex and the replicationfork-associatedfactor,Csm3,arebothimportantmediatorsofpericentromericcohesion,buttheyactthrough distinctmechanisms.WeshowthatcomponentsoftheCtf19complexdirecttheincreasedassociationofcohesinwiththe pericentromere.Incontrast,Csm3isdispensableforcohesinenrichmentinthepericentromerebutisessentialinensuring itsfunctionalityinholdingsistercentromerestogether.Consistently,cellslackingCsm3showadditivecohesiondefectsin combination with mutants in the Ctf19 complex. Furthermore, delaying DNA replication rescues the cohesion defect observedincellslackingCtf19complexcomponents,butnotCsm3.WeproposethattheCtf19complexensuresadditional loadingofcohesinatcentromerespriortopassageofthereplicationfork,therebyensuringitsincorporationintofunctional linkagesthroughaprocessrequiringCsm3.
