It is not known whether this defect may favor lung colonization by opportunisticpathogens. PrincipalFindings:WehaveanalyzedtheeffectsofextracellularGSHontheabilityofBurkholderiacenocepaciatopenetrate andmultiplyinepithelialrespiratorycells.ExtracellularGSHprovedtobeabletodrasticallyreducethepathogenabilityto adhereandinvadeairwayepithelialcells.ThiseffectiscorrelatedtoaGSH-dependentincreaseinthenumberoffreethiols onthesurfaceofepithelialcells,suggestiveofachangeintheoxidoreductivestatusofmembraneproteinsinvolvedinB. cenocepaciarecognition.Moreover,treatmentswithGSHledtoaconsistentreductionoftheexpressionofIL-8,TNF-aand IL-1binresponsetoB.cenocepaciainfection. Conclusions and Significance: Extracellular GSH modulates the interaction between B. cenocepacia and epithelial respiratorycellsandinhibitsthebacterialinvasionintothesecells.Thissuggeststhattherapiesaimedatrestoringnormal levelsofGSHintheASLmightbebeneficialtocontrolCFlunginfections.
