OriginandEvolutionofDengueVirusType3inBrazil Jose′lioMariaGalva?odeArau′jo1*.,GonzaloBello2.,HectorRomero3,RitaMariaRibeiroNogueira4 1Laboratory of Molecular Biology for Infectious Diseases and Cancer, Federal University of Rio Grande do Norte, Natal, Brazil, 2Laboratorio de AIDS e Imunologia Molecular,InstitutoOswaldoCruz–FIOCRUZ,RiodeJaneiro,Brazil,3LaboratoriodeOrganizacio′nyEvolucio′ndelGenoma,Seccio′nBiomatema′ticas,FacultaddeCiencias, UniversidaddelaRepu′blica,Montevideo,Uruguay,4LaboratoryofFlavivirus,OswaldoCruzInstitute–FIOCRUZ,RiodeJaneiro,Brazil Abstract The incidence of dengue fever and dengue hemorrhagic fever in Brazil experienced a significant increase since the emergenceofdenguevirustype-3(DENV-3)attheearly2000s.Despitethemajorpublichealthconcerns,therehavebeen veryfewstudiesofthemolecularepidemiologyandtime-scaleofthisDENVlineageinBrazil.Inthisstudy,weinvestigated the origin and dispersion dynamics of DENV-3 genotype III in Brazil by examining a large number (n=107) of E gene sequencessampledbetween2001and2009fromdiverseBrazilianregions.TheseBraziliansequenceswerecombinedwith 457DENV-3genotypeIIIEgenesequencesfrom29countriesaroundtheworld.Ourphylogeneticanalysisrevealsthatthere have been at least four introductions of the DENV-3 genotype III in Brazil, as signified by the presence of four phylogenetically distinct lineages.Threelineages(BR-I, BR-II, andBR-III) wereprobably imported fromtheLesserAntilles (Caribbean),whilethefourthone(BR-IV)wasprobablyintroducedfromColombiaorVenezuela.WhilelineagesBR-IandBR- IIsucceededingettingestablishedanddisseminatedinBrazilandothercountriesfromtheSouthernCone,lineagesBR-III andBR-IVwereonlydetectedinonesingleindividualeachfromtheNorthregion.Thephylogeographicanalysisindicates thatDENV-3lineagesBR-IandBR-IIweremostlikelyintroducedintoBrazilthroughtheSoutheastandNorthregionsaround 1999(95%HPD:1998–2000)and2001(95%HPD:2000–2002),respectively.ThesefindingsshowthatimportationofDENV-3 lineagesfromtheCaribbeanislands
