AlthoughPALS1isacentralcomponentofthecellpolaritynetwork,itsexpressionandfunctioninlymphocytesremains unknown.HereweinvestigatedwhetherPALS1ispresentinTcellsandwhetheritcontributestoTCell-Receptor(TCR)- mediatedactivation. Methodology/Principal Findings: By combining RT-PCR and immunoblot assays, we found that PALS1 is constitutively expressedinhumanTlymphocytesaswellasinJurkatTcells.siRNA-basedknockdownofPALS1hamperedTCR-induced activationandoptimalproliferationoflymphocyte.WefurtherprovideevidencethatPALS1depletionselectivelyhindered TCR-drivenactivationofthetranscriptionfactorNF-kB. Conclusions:ThecellpolarityproteinPALS1isexpressedinTlymphocytesandparticipatestotheoptimalactivationofNF- kBfollowingTCRstimulation.
