Tambourgi1* 1ImmunochemistryLaboratory,ButantanInstitute,Sa?o Paulo,Sa?o Paulo,Brazil,2GeneticsLaboratory,ButantanInstitute,Sa?o Paulo,Sa?o Paulo,Brazil,3Departmentof Pharmacology,OncologyandRadiology,SchoolofMedicine,CardiffUniversity,Cardiff,UnitedKingdom Abstract Background:ThecaterpillarofthemothPremolissemirufa(Lepidoptera:Arctiidae),commonlynamedPararama,isendemic oftheAmazonbasin.Accidentalcontactwiththesecaterpillarbristlescauseslocalsymptomssuchasintenseheat,pain, edema and itching which last for three to seven days; however, after multiples contacts, it may induce joint-space narrowingandbonealteration,aswellasdegenerationofthearticularcartilageandimmobilizationoftheaffectedjoints. Specific treatment for this disease does not exist, but corticosteroids are frequently administered. Despite of the public healthhazardofPremolissemirufacaterpillarpoisoning,littleisknownaboutthenatureofthetoxiccomponentsinvolved intheinductionofthepathology. Methodology/Principal Findings: Here we have investigated the biological and immunochemical characteristics of the caterpillar’sbristlescomponents.Analysisofthebristlesextractininvitroassaysrevealedthepresenceofproteolyticand hyaluronidaseactivitiesbutnophospholipaseA2activity.Invivo,itwasobservedthatthebristlesextractisnotlethalbut caninduceanintenseinflammatoryprocess,characterizedbythepresenceofneutrophilsinthepawtissuesofinjected mice.Furthermore,thebristlescomponentsstimulatedanintenseandspecificantibodyresponsebutautoantibodiessuch asanti-DNAoranti-collagentypeIIwerenotdetected. Conclusion:TheresultssuggestthatPremolissemirufacaterpillarbristlessecretioncontainsamixtureofdifferentenzymes thatmay acttogether i
