The bovine prion gene (PRNP) contains regions of both high and low linkage disequilibrium (LD) that appear to be conserved across Bos tauruspopulations.TheregionofhighLD,whichspansthepromoterandpartofintron2,containspolymorphiclocithat have been associated with classical BSE status. However, the complex genetic architecture of PRNP has not been systematicallytestedforanassociationwithclassicalBSE. Methodology/PrincipalFindings:Inthisstudy,haplotypetaggingsinglenucleotidepolymorphisms(htSNPs)withinPRNP wereusedtotestforassociationbetweenPRNPhaplotypesandBSEdisease.AcombinationofIlluminagoldengateassay, sequencingandPCRamplificationwasusedtogenotype18htSNPsand2indelsin95BSEcaseand134controlanimals.A haplotypewithintheregionofhighLDwasfoundtobeassociatedwithBSEunaffectedanimals(p-value=0.000114). Conclusion/Significance:APRNPhaplotypeassociationwithclassicalBSEincidencehasbeenidentified.Thisresultsuggests thatageneticdeterminantinornearPRNPmayinfluenceclassicalBSEincidenceincattle. Citation:
