Methods:Serumresistinlevelsweredeterminedin42patientswithinflammatorymyopathiesand27healthy controls.Theassociationamongresistinlevels,inflammation,globaldiseaseactivityandmusclestrengthwas examined.Theexpressionofresistininmuscletissuesfrompatientswithinflammatorymyopathiesandhealthy controlswasevaluated.Geneexpressionandproteinreleasefromresistin-stimulatedmuscleandmononuclearcells wereassessed. Results:Inpatientswithinflammatorymyopathies,theserumlevelsofresistinweresignificantlyhigherthanthose observedincontrols(8.53±6.84vs.4.54±1.08ng/ml,P0.0001)andcorrelatedwithC-reactiveprotein(CRP) levels(r=0.328,P=0.044)andmyositisdiseaseactivityassessmentvisualanaloguescales(MYOACT)(r=0.382,P =0.026).StrongerassociationwasobservedbetweenthelevelsofserumresistinandCRPlevels(r=0.717,P= 0.037)aswellasMYOACT(r=0.798,P=0.007),andtherewasatrendtowardscorrelationbetweenserumresistin andmyoglobinlevels(r=0.650,P=0.067)inanti-Jo-1positivepatients.Furthermore,inpatientswith dermatomyositis,serumresistinlevelssignificantlycorrelatedwithMYOACT(r=0.667,P=0.001),creatinekinase(r =0.739,P=0.001)andmyoglobinlevels(r=0.791,P=0.0003)andshowedatrendtowardscorrelationwithCRP levels(r=0.447,P=0.067).Resistinexpressioninmuscletissuewassignificantlyhigherinpatientswith inflammatorymyopathiescomparedtocontrols,andresistininducedtheexpressionofinterleukins(IL)-1bandIL-6 andmonocytechemoattractantprotein(MCP)-1inmononuclearcellsbutnotinmyocytes.
