Resting-StateFunctionalConnectivityBiasofMiddle TemporalGyrusandCaudatewithAlteredGrayMatter VolumeinMajorDepression ChaoqiongMa1,JurongDing1,JunLi1,WenbinGuo2,3,ZhiliangLong1,FengLiu1,QingGao1,LingZeng1, JingpingZhao2*,HuafuChen1* 1Key Laboratory for NeuroInformation of Ministry of Education, School of Life Science and Technology, University of Electronic Science and Technology of China, Chengdu,PRChina,2MentalHealthInstitute,TheSecondXiangyaHospital,CentralSouthUniversityChangsha,Hunan,China, 3MentalHealthCenter,FirstAffiliated Hospital,GuangxiMedicalUniversity,Nanning,Guangxi,China Abstract Magnetic resonance imaging (MRI) studies have indicated that the structure deficits and resting-state functional connectivity(FC)imbalancesincortico-limbiccircuitrymightunderlinethepathophysiologyofMDD.Usingstructureand functionalMRI,ouraimistoinvestigategraymatterabnormalitiesinpatientswithtreatment-resistantdepression(TRD)and treatment-responsivedepression(TSD),andtestwhetherthealteredgraymatterisassociatedwithalteredFC.Voxel-based morphometry was used to investigate the regions with gray matter abnormality and FC analysis was further conducted betweeneachgraymatterabnormalregionandtheremainingvoxelsinthebrain.Usingone-wayanalysisofvariance,we foundsignificantgraymatterabnormalitiesintherightmiddletemporalcortex(MTG)andbilateralcaudateamongtheTRD, TSDandhealthycontrols.FortheFCoftherightMTG,wefoundthatboththepatientswithTRDandTSDshowedaltered connectivity mainly in the default-mode network (DMN). For the FC of the right caudate, both patient groups showed alteredconnectivityinthefrontalregions.OurresultsrevealedthegraymatterreductionofrightMTGandbilateralcaudate, anddisruptedfunctionalconnectiontowidelydistributedcircuitryinDMNandfrontalregions,respectively.Theseresults suggestthattheabnormalDMNandrewardcircuitactivitymightbebiomarkersofdepressiontrait.
