WefoundthatthemRNAsencodingCdc42,RhoQ,Rac1, Rac2,RhoAandRhoCarethemostabundant.Inaddition,RhoG,RhoB,RhoFandRhoVareexpressedatlowlevelsoronlyin specificcelltypes.Moredifferentiatedcellsalongthemonocyte-lineagedisplaylowerlevelsofCdc42andRhoV,whileRhoC mRNAismoreabundant.Inaddition,theRhoGTPaseexpressionprofilechangesduringdendriticcellmaturationwithRac1 beingupregulatedandRac2downregulated.Finally,GM-CSFstimulation,duringmacrophageandosteoclastdifferentiation, leadstohighexpressionofRac2,whileM-CSFinduceshighlevelsofRhoA,showingthatthesecytokinesinduceadistinct pattern.OurdatauncovercelltypespecificmodulationoftheRhoGTPaseexpressionprofileinhematopoieticstemcells andinmoredifferentiatedcellsofthemyeloidlineage.
