However, mechanistic data on these small molecules in atherosclerosis (AS) are still missing. The presentstudyaimstoinvestigatethepotentialroleofmiRNAsinAS. MethodsandResults:ThemiRNAtranscriptasewasverifiedbyTaqManreal-timepolymerasechainreactionassay.Thoracic aortasampleswereobtainedfromApolipoproteinEknockoutmice,andplasmasampleswerefromcoronaryarterydisease (CAD)patients.TheresultsshowedthatthemiR-155levelwasthemostsignificantlyelevatedbothinASmiceandCAD patientsrelativetothenormalcontrol.ThefunctionalroleofmiR-155intheatheroscleroticpathphysiologicalprocesswas alsoobservedinvivoandinvitro.TheobservationssuggestedthatmiR-155isapartofanegativefeedbackloop,which down-modulatesinflammatorycytokineproductionanddecreasesASprogression.miR-155wasalsofoundtomediatethe inflammatory response and mitogen-activated protein kinase (MAPK) pathway by targeting mitogen-activated protein kinasekinasekinase10.
