Methods:Single,doubleandtripleimmunohistochemistryaswellasimmunofluorescencestainingwasperformed onminorsalivaryglandtissueretrievedfrompSS,chronicallyinflamedandnormalsubjects. Results:WedetectedsignificantnumbersofCD138+,non-proliferating,Bcl-2expressingplasmacellsinthesalivary glandsofpSSpatientswithhighfocusscore(FS).Furthermore,wedemonstratedthatCXCL12andinterleukin(IL)-6 survivalfactorswerehighlyexpressedinpSSsalivaryglandepitheliumandbyfocalmononuclearinfiltratingcells. Notably,adipocyteswhenpresentinthesalivaryglandtissuewereanimportantsourceofCXCL12.Weclearly demonstratethatplasmacellsarelocalisedincloseproximitytoCXCL12andIL-6expressingcellsandthusthat theenvironmentofsalivaryglandswithhighFSprovidefactorsvitalforplasmacellsurvival. Conclusions:PlasmacellsresidinginthesalivaryglandsofpSSpatientswithhighFSshowedphenotypic characteristicsofthelong-livedplasmacellsubtype.Furthermore,thepSSsalivaryglandmicroenvironment providednichesrichinfactorsvitalforplasmacellsurvival.
