Advancement in cancer research has demonstrated the potential roleofvarious tumorsuppressor proteinsin inhibitionofcancer progression. Currentstudies haveshownthat axonalsproutinginhibitor,semaphorin3A(Sema3A)actsasapotentsuppressoroftumorangiogenesisinvariouscancer models.However,thefunctionofSema3Ainregulationofmelanomaprogressionisnotwellstudied,andyettobethe subjectofintenseinvestigation. Methodology/PrincipalFindings:Inthisstudy,usingmultipleinvitroandinvivoapproacheswehavedemonstratedthat Sema3Aactsasapotenttumorsuppressorinvitroandinvivomice(C57BL/6)models.Mousemelanoma(B16F10)cells overexpressed with Sema 3A resulted in significant inhibition of cell motility, invasiveness and proliferation as well as suppressionofinvivotumorgrowth,angiogenesisandmetastasisinmicemodels.Moreover,wehaveobservedthatSema 3AoverexpressedmelanomacloneshowedincreasedsensitivitytowardscurcuminandDacarbazine,anti-canceragents. Conclusions:Ourresultsdemonstrate,atleastinpart,thefunctionalapproachunderlyingSema3Amediatedinhibitionof tumorigenesis and angiogenesis and a clear understanding of such a process may facilitate the development of novel therapeuticstrategyforthetreatmentofcancer.
