ExtensivestudiesonmiRNAshavebeenperformedindifferentplantssuchasArabidopsisthaliana,Oryzasativaetc. andvolumeofthemiRNAdatabase,mirBASE,hasbeenincreasingondaytodaybasis.SteviarebaudianaBertoniis animportantperennialherbwhichaccumulateshighconcentrationsofditerpenesteviolglycosideswhich contributestoitshighindexedsweeteningpropertywithnocalorificvalue.Severalstudieshavebeencarriedout forunderstandingmolecularmechanisminvolvedinbiosynthesisoftheseglycosides,however,informationabout miRNAshasbeenlackinginS.rebaudiana.DeepsequencingofsmallRNAscombinedwithtranscriptomicdataisa powerfultoolforidentifyingconservedandnovelmiRNAsirrespectiveofavailabilityofgenomesequencedata. Results:ToidentifymiRNAsinS.rebaudiana,sRNAlibrarywasconstructedandsequencedusingIlluminagenome analyzerII.Atotalof30,472,534readsrepresenting2,509,190distinctsequenceswereobtainedfromsRNAlibrary. Basedonsequencesimilarity,weidentified100miRNAsbelongingto34highlyconservedfamilies.Also,we identified12novelmiRNAswhoseprecursorswerepotentiallygeneratedfromsteviaESTandnucleotide sequences.Allnovelsequenceshavenotbeenearlierdescribedinotherplantspecies.Putativetargetgeneswere predictedformostconservedandnovelmiRNAs.ThepredictedtargetsaremainlymRNAencodingenzymes regulatingessentialplantmetabolicandsignalingpathways. Conclusions:Thisstudyledtotheidentificationof34highlyconservedmiRNAfamiliesand12novelpotential miRNAsindicatingthatspecificmiRNAsexistinsteviaspecies.OurresultsprovidedinformationonsteviamiRNAs andtheirtargetsbuildingafoundationforfuturestudiestounderstandtheirrolesinkeysteviatraits.
