Smyd3IsRequiredfortheDevelopmentofCardiacand SkeletalMuscleinZebrafish TomoakiFujii1,Shin-ichiroTsunesumi1,KiyoshiYamaguchi1,SumikoWatanabe2,YoichiFurukawa1* 1Division of Clinical Genome Research, Advanced Clinical Research Center, Institute of Medical Science, The University of Tokyo, Japan, 2Division of Molecular DevelopmentalBiology,InstituteofMedicalScience,TheUniversityofTokyo,Tokyo,Japan Abstract Modificationsofhistonetailsareinvolvedintheregulationofawiderangeofbiologicalprocessesincludingcellcycle,cell survival,celldivision,andcelldifferentiation.Amongthemodifications,histonemethylationplaysacriticalroleincardiac andskeletalmuscledifferentiation.Inourearlierstudies,wefoundthatSMYD3hasmethyltransferaseactivitytohistoneH3 lysine4,andthatitsup-regulationisinvolvedinthetumorigenesisofhumancolon,liver,andbreast.Toclarifytheroleof Smyd3indevelopment,wehavestudieditsexpressionpatternsinzebrafishembryosandtheeffectofitssuppressionon developmentusingSmyd3-specificantisensemorpholino-oligonucleotides.Wehereshowthattranscriptsofsmyd3were expressedinzebrafishembryosatalldevelopmentalstagesexaminedandthatknockdownofsmyd3inembryosresultedin pericardial edema and defects in the trunk structure. In addition, these phenotypes were associated with abnormal expression of three heart-chamber markers including cmlc2, amhc and vmhc, and abnormal expression of myogenic regulatoryfactorsincludingmyodandmyog.ThesedatasuggestthatSmyd3playsanimportantroleinthedevelopmentof heartandskeletalmuscle.
