Gene amplifications, a known activatingmechanismofoncogenes,targetthe3q26-qterregionasoneofthemostfrequentlygained/amplifiedgenomic sitesinSCCofvarioustypes.Here,weusedarraycomparativegenomichybridizationtodelineatetheconsensusregionof 3q26.3amplificationsinlungSCC.Recurrentamplificationsoccurin20%oflungSCC(136tumorsintotal)andmaptoacore regionof2Mb(Megabases)thatencompassesSOX2,atranscriptionfactorgene.IntenseSOX2immunostainingisfrequent innucleioflungSCC,indicatingpotentialactivetranscriptionalregulationbySOX2.Analysesofthetranscriptomeoflung SCC,SOX2-overexpressinglungepithelialcellsandembryonicstemcells(ESCs)revealthatSOX2contributestoactivateESC- likephenotypesandprovidecluespertainingtothederegulatedgenesinvolvedinthemalignantphenotype.Incellcultu
