However,thefunctionalroleofantisensetranscriptshasbeendemonstratedonlyinafewcasesandithasbeen suggestedthatmostantisensetranscriptsmayresultfrompromiscuousbi-directionaltranscriptioninadense genome. Results:Here,weusestrand-specificRNAsequencingtostudyanti-sensetranscriptioninSaccharomycescerevisiae. Wedetect1,103putativeantisensetranscriptsexpressedinmid-logphasegrowth,rangingfrom39short transcriptscoveringonlythe3’UTRofsensegenesto145longtranscriptscoveringtheentiresenseopenreading frame.Manyoftheseantisensetranscriptsoverlapsensegenesthatarerepressedinmid-logphaseandare importantinstationaryphase,stressresponse,ormeiosis.Wevalidatethedifferentialregulationof67antisense transcriptsandtheirsensetargetsinrelevantconditions,includingnutrientlimitationandenvironmentalstresses. Moreover,weshowthatseveralantisensetranscriptsand,insomecases,theirdifferentialexpressionhavebeen conservedacrossfivespeciesofyeastspanning150millionyearsofevolution.Divergenceintheregulationof antisensetranscriptstotworespiratorygenescoincideswiththeevolutionofrespiro-fermentation.
