Methods:Thisarticledescribesconcepts,methodsandprocedurestoassistinthedesignofanimalandtissue/cell basedstudiesofsexdifferencesincardiovascularstructure,functionandmodelsofdisease. Results:Toaddressknowledgegaps,studydesignsmustincorporateappropriateexperimentalmaterialincluding species/straincharacteristics,sexandhormonalstatus.Determiningwhetherasexdifferenceexistsinatraitmust takeintoaccountthereproductivestatusandhistoryoftheanimalincludingthoseusedfortissue(cell)harvest, suchasthepresenceofgonadalsteroidsatthetimeoftesting,duringdevelopmentornumberofpregnancies. Whenselectingthetypeofexperimentalanimal,additionalconsiderationshouldbegiventodietrequirements (soyorplantbasedinfluencingconsumptionofphytoestrogen),lifespan,frequencyofestrouscycleinfemales,and abilitytoinvestigatedevelopmentalorenvironmentalcomponentsofdiseasemodulation.Stressimposedby disruptionofsleep/wakecycles,patternsofsocialinteraction(ordegreeofsocialisolation),orhandlingmay influenceadrenalhormonesthatinteractwithpathwaysactivatedbythesexsteroidhormones.Caremustbe giventoselectionofhormonaltreatmentandrouteofadministration.
