However,frequentandseriousadverseeffectsofNSAIDshavebeenrecognized.Intra-articularinjectionsofhyaluronic acid(IA-HA)forthetreatmentofkneeOAhavebeenshowntoreducepainandimprovejointfunction.However,there hasbeennoqualifieddirectcomparisonstudyoftheefficacyandsafetybetweenIA-HAandNSAIDsforpatientswith kneeOA.Theaimofthisstudywastoclarifytheefficacyandsafetyofearly-phaseIA-HAincomparisontothoseof NSAIDsforpatientswithkneeOA. Methods:Thismulticenter,randomized,open-label,parallel-group,non-inferioritycomparisonstudywithanoralNSAID involvedatotalof200patientswithkneeOA.Anindependent,computer-generatedrandomizationsequencewasused torandomlyassignpatientsina1:1ratiotoNSAIDsthreetimesperdayforfiveweeks(n=100)orIA-HAonceaweekfor fiveweeks(n=100).Theprimaryendpointwasthepercentagechangeinthepatient-orientedoutcomemeasureforknee OA,theJapaneseKneeOsteoarthritisMeasure(JKOM)score.Allpatientswerequestionedregardinganyadverseevents duringtreatment.Thefullanalysisset(FAS)wasusedforanalysis.Themarginofnon-inferioritywas10%.
