Methods Real-time quantitative PCR (RT-qPCR) method and immunohistochemistry methods was used to detect the expression of Wnt2 in HCC, paired Paracancerous tissue and normal liver tissue;RNAi was used to knock down the expression of Wnt2 in HepG2,and the change of Wnt2 were detected by RT-qPCR and Western blot,and testing RNAi cell proliferation level and cycle change. Result (1).Wnt2 expressed in the highest in cancer, paired Paracancerous tissue is the second smallest,and normal liver minimum. (2). Using RNAi technology,The expression of Wnt2 was downregulated.HepG2 proliferation was inhibition and the cell growth mainly detained in G0/G1 and the percentage of S decreased. Conclusion The expression of Wnt2 Was higher in tumor tissues than in normal liver tissues;Using RNAi technology efficiently knocks down the expression of Wnt2 in Hepatocellular cell line HepG2,results in suppression of tumor cell growth.siRNA against Wnt2 is a potential therapeutic target for Human Hepatocellular Carcinoma.
