应用组织芯片技术探究结直肠癌中Lumican表达及意义 　 作者：宋洪江，佟丹丹，金银姬 孟祥宁，白静，宋春芳 【摘要】 ［目的］ 探讨Lumican的表达与结直肠癌发生、发展的关系。［方法］ 构建100对结直肠癌及相应癌旁正常黏膜上皮组织的组织芯片，应用免疫组化方法检测Lumican的表达强度。［结果］ Lumican在结直肠癌组织中的表达率（84.0％）显著高于癌旁正常黏膜中的表达率（42.6％）（Plt;0.01）。Lumican在腺癌中的表达率（87.2％）高于在黏液腺癌及印戒细胞癌中的表达率（68.8％）(Plt;0.05)。Lumican的表达强度与结直肠癌浸润深度之间存在非常显著的正相关（r=0.07959，Plt;0.01）及线性关系(χ2=5.2148，Plt;0.05)，即随着浸润程度加深，Lumican的表达强度逐渐增加。Lumican的表达强度与患者的年龄、性别、发病部位、淋巴结是否转移及分化程度无关（Pgt;0.05）。［结论］ Lumican的异常表达可能是结直肠癌发生的早期事件，与结直肠癌的组织分型和侵袭能力有关，在结直肠癌的发生发展中发挥重要作用。 【关键词】 肠肿瘤 Application of Tissue Chip Assay to Expression of Lumican for Colorectal Cancer and its Significance Abstract: ［Purpose］To explore the relationship of Lumican expression with carcinogenesis and progression in colorectal cancer. ［Methods］ One hundred couples of tissue chips of colorectal cancer tissues and adjacent normal epithelium mucosa tissues were constructed. Expression intensity of Lumincan was measured by immunohistochemical method. ［Results］ Lumican expression ratio in colorectal cancer tissues (84.0%) was significantly higher than that (42.6%) in adjacent normal epithelium mucosa tissues (Plt;0.01). Lumican expression ratio in adenocarcinoma (87.2%) was higher than that in mucinous adenocarcinoma and signet-ring cell carcinoma (68.8%), (Plt;0.05). There was a distinctly positive correlation between intensity of Lumican expression and invasive depth in colorectal cancer (r=0.07959,Plt;0.01), as well as linear correlation (χ2=5.2148，Plt;0.05). Lumican expression intensity increased as invasive depth increased in colorectal cancer. Lumican expression intensity didn’t relate to age, sex, tumor location, lymph node metastasis and cell differentiation (Pgt;0.05). ［Conclusion］ Mal-expression of Lumican correlates to histopathologic type and invasive capability in colorectal cancer.
