Abstract:Purpose To investigat the extraction process conditions of polysaccharide from yeast.Methods: Yeast was extracted by ethanol and then extract was decolorized by activive carbon and purifieded using the ion exchange resins.The eluent was crystallized by alchol. Results:yeast was extracted with 70% ethanol solution at 70℃ for 1.5h,and decolcrized with 1% active carbon at 50℃ and pH 4.8,then purified by 711 and 122 ion exchange resins .The final yield of polysaccharide was 11% with purity of 98%. Method: The optimal conditions for immobilization were obtained through orthogonal experiments and the synthesis conditions were optimized with the conversion rate as an index. Study on Synthesis of Salidroside Using Immobilization β- glucosidase Result : The highest recovery activity of immobilization β- glucosidase (74.38 %) was obtained with chitosan concentration of 1.5 % , adsorption time of 9 hours , cross-linking time of 12h, glutaraldehyde concentration of 1.0% and adsorption temperature of 0℃. The research on synthesis reaction conditions also showed that the conversion rate of salidroside could reach 71.9% at the pH 5.8 acetate buffer/tert-butyl alcohol (1:9) system when reaction time was 50 hours ,reaction temperature was 50℃, tyrosol concentration was 5g/L and substrate molar ratio
