AbstractIt is well known that pancreatic cancer is one of the malignant tumors that has the worst therapeutic effect, commonly used treatments include surgical, radiotherapy, chemotherapy and molecular targeted therapy. DNA sequencer-assisted fluorophore-assisted carbohydrate electrophoresis (DSA-FACE) has been introduced as a robust, high-throughput, exceedingly sensitive, and reliably quantitative method, which has the advantages of efficient, high-throughput have become the important technology in the study of glycobiology. At present, the DSA-FACE technology usually been used to the study of the serum samples and in the tissue samples have not been reported.This experiment have been optimized the DSA-FACE technology for the first time and applied the DSA-FACE technology to the pancreatic cancer tissue samples, successfully established a technology of the cell surface N-glycans detection of pancreatic carcinoma. On this basis, we has investigated the relationship between human N-glycan profiles and age, gender by DSA-FACE. We analysis 41 pairs of pancreatic carcinoma tissue cell surface N-liycans. Compared with paraneoplastic group, cancer group with peak 10, peak 13 were significantly increased; peak 32, peak 40 were significantly decreased. In addition, we compared different group between gender, age and pathological. Peak 10 relates to age factor and is significantly increased in cancer group with more than 60 years old patients; peak 32 and peak 40 are significantly increased in paraneoplastic group with male patients and sharply decline when cancer.
