4.TheexpressionsofTRPC6,CaNmRNAintherenalcortexinnaturalprogression andundertheinterventionofFK506were detectedusingReal-time Ploymerase Chain Reaction;5.TheexpressionsanddistributionsofTRPC6,CaNproteinsinratrenal tissuesweredetectedbyimmunohistochemicaltechniques;6. The expressionsof TRPC6, CaNproteinsin therenalcortex were detected by western blot.Results:Afterthefirstinjectionofadriamycin,asignificantincreasementofthe24-hour urinaryprotein inadriamycinratswasobserved at3rd weekandpersistedupto7thweek（P＜0.01），theadriamycinratsshowedfattysubstancemetabolicdisorder，hypoproteinemiaandrenalfunctionallesionwhicharetypicalperformancesof nephroticsyndrome.ComparedwithADRrats,FK506couldalleviatehyperlipemia, hypoproteinemiaandrenalinsufficiencyto some extent.Theadriamycinratsdevelopedfromminimalchangediseasetofocalsegmental glomerulosclerosisfom3rdto7thweekobservedbylightmicroscopeandthefoot processesbroadenedtoadifferentextentat3rdweek, thediffusefusionand effacementeven disappearoffootprocesseswereobservedat7thweekby transmission electronmicroscope.ComparedwithADRrats,FK506couldreducefootprocessesfusionsubepithelial electron densedepositsobviously.Comparedwiththecontrolrats,mRNAexpressionofTRPC6inadriamycinrats begantoincreaseatweek3(P＜0.05)andpersisteduptoweek7(P＜0.01),mRNA expressionofCaNinadriamycinratswasincreased(P＜0.05~0.01)atanyobserved time point.Innormalratrenal, TRPC6washighlyexpressedintheglomerularcapillary loops,mesangiumarea,podocytesandrenaltubularepithelialcellcytoplasm,and therewaslikelyafewexpressioninthemedulla;CaNwasmainlydistributedin glomerulus,weaklyexpressedintubulointerstitium, andperivascularconnective tissuewasalsoseenpositiveexpression.ThespecificproteinbandsofTRPC6and CaN were detected with the size of 106 and 62 kDa, respectively.At7thweekcomparedwiththeADRrats,mRNA andprotein expressionsofTRPC6andCaNinADR+FK0.5andADR+FK1.0ratsweresignificantlydecreased (P＜0.05~0.01)，expressionsofTRPC6andCaNbetweenADR+FK0.5and ADR+FK1.0 ratsshowed nosigni
