ABSTRACTObjective：ToinvestigatecorrelationbetweenJAK2V617Fgenemutation，CALRgenemutationandclinicalfeature,laboratoryexaminationofBCR/ABLnegativePV,ET,PMFpatients.Materialsandmethods：Thereareeighty-oneBCR/ABLnegativePV,ET,PMFpatients,37polycythemiavera(PV),26secondarypolycythemiapatientswhosehaemoglobincountnearly160g/L,39essentialthrombocythemia(ET),28secondarythrombocythemiapatientswhoseplateletcountnearly450×109/L,5primarymyelofibrosis(PMF),5secondarymyelofibrosis.JAK2V617FgenemutationwasdetectedbyAS-PCRmethod,CALRgenemutationwasdetectedbySANGERmethod,toinvestigatewhetherthedifferentcharacteristics,laboratoryexaminationisrelatedwithgenemutation.Results:1.59/81BCR/ABLnegativePV,ET,PMFpatientsdetectedJAK2V617Fgenemutation,thepositiveexpressionratewas61.53%(16/26)inPVpatients,%，16/28)inETpatientsand40.0%(2/5)inPMFpatients.Inthecontrolgroup,Theother32patientswerenegative.ThereweresignificantdifferencesintheJAK2V617FgenemutationbetweenPV,ETandsecondarydiseases,sotodetectithasclinicalvaluetodiscriminatethesediseases.TherewasnodifferencebetweenPMFandsecondarymyelofibrosis.TheincidenceofPVpatients,leucocytecount,haemoglobin,plateletcount,neutrophilegranulocytecountwashigherthansecondarypolycythemia(P0.05);TheincidenceoccurredofETpatients,leucocytecount,haemoglobinlevel,andsplenomegalywasgreaterthansecondarythrombocythemia（P0.05）;TheincidenceofPMFpatients,plateletcount,neutrophilegranulocytecountwashigherthansecondarymyelofibrosis(P0.05).TheincidenceofPVpatientswithJAK2V617Fgenemutationpositive,leucocytecount,haemoglobin,neutrophilegranulocytecountwashigherthanJAK2V617FnegativePVpatients;TheincidenceofETpatientswithJAK2V617Fgenemutationpositive,leucocytecount,haemoglobin,neutrophilegranulocytecountwashigherthannegativegroup;TheincidenceofinPMFpatientswithJAK2V617Fgenemutationpositive,leucocytecount，neutrophilegranulocytecountwashigherthannegatibvegroup.ThetotalincidenceofthrombosisinJAK2V617Fgenemutationpatientswashigherthannegativepatients.TheincidenceofETpatientswithCALR
