The expression of Sipa1 was detected by immunohistochemistry.Five SNPs of Sipa1 gene were explorded by PCR-LDR method: 383 cases of gastric carcinoma after surgery opertation in the First Affiliated Hospital of Fujian Medical University from 2001 to 2009 were collected, 185 health contorls were recruited form routine check-up people who had no cancer history or family cancer predispositions. Genomic DNA was extracted from the histologically normal formalin-fixed paraffin-embedded tissues in the distant margin to the gastric cancer and fresh blood of the controls. PCR-LDR and sequencing were used to determine thegenotypes and allele frenquency of rs931127（-313A/G）、rs3741379（316GT）、rs3741378（545CT）、rs2509948（-92+740CT）and rs2448490（985-391GA）of Sipa1 gene.Results:The expression of Sipa1 protein in gastric intestial metaplasia mucosa was higher than in normal gastric mucosa.The expression of Sipa1 protein at normal gastric mucosa tissuses was less than in cancer cells of gastric mucosa.The expression of Sipa1 protein in well-differentiated-moderately-differentiated adenocarcinoma and intestinal type gastric carcinoma is higher than inmoderately-differentiated-poorly-differentiated adenocarcinoma 、 mucousadenocarcinoma、singnet-ring cell carcinoma 、diffused type gastric carcinoma.From the gastric mucosa layer to the invasiveness frontier, the expression of Sipa1 protein showed downtrend, and from the invasiveness frontier to the metastsis lymph nodes, the expression of Sipa1 protein showed accending tendency.The expression of Sipa1 protein is negatively related with the survival time of patients after operation.There was no difference of distribution frequency of Sipa1 rs931127（-313A/G）、rs3741379（316GT）、rs3741378（545CT）、rs2509948（-92+740CT） andrs2448490（985-391GA）genotypes in the gastric carcinoma patients from controls.Conclusions:The expression of Sipa1 protein is involved in the tissue differentiation and the invasiveness and metastasis, moreover, it is ne
