Expressionandregulationstudyonsilkworm(Bombyxmori)β-N-AcetylglucosaminidasesAbstractExpressionandregulationstudyonsilkworm(Bombyxmori)β-N-AcetylglucosaminidasesAbstractDuringthemoltingprocess,insectscouldn’tgrowanddevelopnormallyifthedegradationandformationofcuticulewasdisturbed.Chitinisanimportantcomponentofinsectcuticle.Sothebiosynthesisanddegradationofchitinarecloselyrelatedwithinsectgrowthanddevelopment.β-N-Acetylglucosaminidase(GlcNAcase)isakeyenzymeinthechitindecompositionprocess.Thesilkworm(Bombyxmori)isnotonlyamodelorganismforlepidopteraninsects,butalsoeconomicallyimportantinsilkindustry.Andtheyarethematerialsofbioreactorandnewinsectindustry.Tofurtherinvestigatethegeneticrelationshipwithotherrelatedenzymesinvolvedinthespecies,espressioninvarioustissuesandpromoteranctivityofsilkwormβ-N-Acetylglucosaminidaseenzymes(BmGlcNAcases),thepresentstudywasaddressed.Themainresultswereshownasfollows:WestudiedtheBmGlcNAcasesusingbioinformaticsmethods.TheresultsshowthatBmGlcNAcase1，BmCHiGlcNAcaseandBmFDLareclosedrelatedwithGlcNAcasesinotherinsects,butBmGlcNAcase2，BmGlcNAcase3，BmGlcNAcase4havecloselyrelationshipwithhexosaminaseinmammalianratherthaninsects.Sequencealignmentshowedtheseproteinshavehighlyconservedcatalyticdomain.Usingdual-spike-inqPCRmethod,weexaminedtheexpressionofBmGlcNAcasesinvarioustissuesofsilkwormaswellasexpressionchangesafterstimulationwithecdysone.TheexpressionlevelsoftheBmGlcNAcasegenesvariedindifferenttissues,andallwerehigher48hafterexposuretoecdysone.Theespressionlevelofallgeneswereincreasedsignificantly(p0.05)exceptBmFDL.WeconcluedthatmoultinghormoneregulatetheexpressionofBmGlcNAcases.AbstractExpressionandregulationstudyonsilkworm(Bombyxmori)β-N-AcetylglucosaminidasesInordertostudythemechanismsunderlyingregulationofBmGlcNAcase1,functionalanalysisofthepromotersofBmGlcNAcase1fromBombyxmoriwasperformedinthisstudy.RecombinantBacmidcontainingseriallytruncatedpromoterfragmentsofBmGlcNAcase1weretransfectedintoSf9cells,thancollectedAcMNPVVirusesfromSf9cells
