中文摘要中文摘要重庆大学硕重庆大学硕士学位论文II PAGE PAGE VI摘 要海洋放线菌次生代谢产物存在着多样和新异的结构类型，这些新的化合物对 于寻找到治疗新疾病的先导化合物是一个重要的资源库。为了寻找具有新的抗菌、 细胞毒活性的天然产物，本文选取了从中国南海珊瑚组织中分离纯化得到的放线 菌作为研究对象，开展了抗菌、抗细胞毒活性次级代谢产物的研究工作，研究内 容主要包括：活性菌株的筛选和发酵条件的考察；代谢产物的分离纯化及结构的 初步鉴定。通过建立以活性筛选为主、化学筛选为辅的二级组合筛选模式，采取简单培 养基初筛、培养基多项选择复筛的步骤，对 56 株从中国南海丛生盔形珊瑚(Galaxea fascicularis)，澄黄滨珊瑚(Porites lutea)，多孔鹿角珊瑚(Acropora millepora)三株珊 瑚中分离出来的海洋放线菌发酵产物进行了生物活性和化学的筛选。对经过初筛 获得的 19 株放线菌进行了抗卤虫复筛，复筛菌株占筛选菌株的比例为：33.9%。综合初筛与复筛的生物活性筛选结果，共获得目标活性菌株 5 株。对 5 株潜在目标活性菌株进行了培养基多项选择复筛工作。通过 HPLC 和 LC-MS 检测的方法， 筛选出次级代谢产物丰富、产量高、活性好的菌株 Streptomyces sp.SCSIO 11469 作 为下一步放大发酵的菌株。对珊瑚共 附生放 线菌 Streptomyces sp.SCSIO 11469 进行放大培 养，采用 HPLC-UV 追踪的方式，利用丙酮、乙酸乙酯等溶剂对发酵产物提取萃取，使用硅 胶、凝胶、反相硅胶等柱色谱分离手段，从该株菌的乙酸乙酯萃取物中分离纯化 得到四个单体化合物(1，2，3，4)。通过理化性质和波谱数据对化合物 3 进行初步的结构鉴定。初步判断化合物 3 为环多肽类化合物。 此外，还对一株来源于南海海洋沉积物的放线菌 Streptomyces sp.SCSIO 11680进行了小规模发酵，利用凝胶、硅胶、中压制备柱等色谱分离纯化技术从该株菌 发酵产物的乙酸乙酯萃取物中分离得到 2 个化合物(5，6)。关键词：珊瑚共附生放线菌，活性筛选，发酵优化，次级代谢产物英文摘要英文摘要重庆大学硕重庆大学硕士学位论文IIIIII PAGE PAGE IVABSTRACTNew-type and diverse of secondary metabolites structure from marine actinomycetes are an important repository to looking for new disease treatment lead compounds. A study on actinomycetes from coral in South China Sea was carried out to explore chemical diversity and search for antibiosis and cytotoxicity metabolites. Studies included screening for microbial strains with antibiosis activities; fermentation studies; isolation, purification and structural elucidation of metabolites.Five antibiosis and cytotoxicity-active actinomycete strains were screened out of fifty-six microbial strains isolated from Galaxea fascicularis 、 Porites lutea andAcropora millepora coral in South China Sea by new gradated-combinatorial screening method.
