III III Functional Analysis of Hamp Domain of ExpS in Dickeya zeae and the Clone of ExpS Domains Yang liping (College of Natural Resources and Environment, South China Agricultural University, Guangzhou 510642, China) Abstract: Rice bacterial foot rot, which is caused by Erwinia chrysanthemi pv.zeae (Ecz), happened in many areas of China and many other countries in Southeast Asia, is one of the important bacterial disease on rice in the past two decades and makes reduction of rice output greatly. At present, very little study is done about its pathogenicity regulation mechanism. Two-component signal transduction systems (TCSs) are a kind of important regulation mechanism and very conservative in many bacteria. It is also an important physiological metabolism control system in Prokaryotes. The two-component systems of ExpS/ExpA has fund in Dickeya zeae. This tstudy revealed that ExpA worked on Dickeya zeae pathogenic on the basis of comprehensive regulation, the further study focus on histidine protein kinase ExpS. Bioinformatics analysis showed that, ExpS consists of 926 amino acids, there are five structural domains, and Hamp is one of it. To study the ExpS domain of the function has important theoretical and practical significance on clarify the pathogenesis of the signal transduction pathway. According to ExpS sequence and domain prediction, designed Hamp, Hk, Hatp, Rec, Hpt 5 functional domains of primers, PCR amplification and received the five structural fragments of clones, and then, expression vectors for further the function of each domain basis. It has great significance for the study of virulence regulation about TCSs in Dickeya zeae, the pathogen of bacterial foot rot disease in rice. In this study, mutant of Hamp domain (△Hamp) from sensor ExpS and its complementary strain were successfully acquired by the way of homologous recombination and marking replacement (gene knock-out).
