AbstractNowadays,thedisposalandcomprehensiveutilizationofwasteactivatedsludge（WAS）hasbecomeaglobalfocus,duetoindustrialandmunicipalwastewatertreatmentplantsproducelargeamountsofWAS.Basedonthelysisofmicrobialcellisakeystepintheprocessofsludgedigest,sludgelysismethodprovidesanewdirectionofresourceutilizationofsludge.Inthisarticle,thefeasibilityofthermophilicmicro-aerobicconditionstoimprovetheperformanceofwasteactivatedsludgelysiswasexplored.Thestudywascarriedoutfromthefollowingaspects:theconditionsofpretreatmentandanalysisofmicrobialcommunity.Thedegradationofmicrobialcellproteinwascombinedwithanalysisofmicrobialcommunitytostudythelysisefficiencyandmechanismofthermophilicmicro-aerobicpretreatmenttosludge.Atfirst,theaerationrateandtemperatureparametersofaffectingsludgelysiswererespectivelyoptimized.Theexperimentgroupreactorwasaeratedgradiently,andtheoptimizedaerationpatternwasobtained.Thereactiontemperaturewasfurtheroptimized,andtheEscherichiacoliwereselectedasatypestraintosimulatethepretreatmentexperiment.TheSDS-polyacrylamidegelelectrophoresis(SDS)wasappliedtoobserveproteindegradationofEscherichiacolibeforeandafterpretreatment.Accordingtotheoptimizedpatternscondition,theactivatedsludgeandEscherichiacoliwererespectivelyscannedbyscanningelectronmicroscopic(SEM)toobservetheeffectofthermophilicmicro-aerobicpretreatmentconditionstostructureandmorphologyofmicrobes.Theresultofstudyshowsthatthepretreatmenteffectofexperimentgroupwasobviouswhentheaerationratewas0.07vvmat70℃for24h.Thesludgelysisofthehighconcentrationoforganicmatterssuchasproteinandtotalsugarwasshown.Thiswasbenefittothelysisofwastesludge,andthebiodegradabilityofwasteactivatedsludgewasobviouslyenhanced.ProteinwasdetectedinsupernatantaftertheEscherichiacoliwerepretreatedinthermophilicmicro-aerobicconditions.AndthecellwallsofEscherichiacoliwerecracked,anditfurtherprovedsthatthermophilicmicro-aerobicpretreatmentforlysisofwastesludgewasfavorable.Thiscouldpromotemacromoleculeorganicmattersthroughcellwalls,andtheywereus
