In ESCC,EGFR gene amplifieation was found in 35 （ 32.71% ） cases,TOPOⅡwas 31 （ 28.97% ） cases.EGFR,TOPOⅡpositive was significantly correlated with histological grades,and no correlation between EGFR or TOPOⅡand gender,age,lymph node or distant metastasis(P0.05).There was a positive correlation between EGFR and TOPOⅡ( r 基 因 = 0.53 0,P0.05). HER2 gene amplifieation was found in 2 （ 1.87% ） cases, and no correlation was obtained between HER2 and gender,age,the depth of invasion, histological grades or lymph node metastasis(P0.05), no relationship between HER2 and EGFR or TOPOⅡ(r 基因 2= 0.23 1,P0.05; r 基因 3=0.169,P0.05). Comparision of EGFR,HER2 and TOPOⅡ protein/gene status detected by IHC, CISH. In ESCC, the general coincident rate of EGFR or TOPOⅡdetected by IHC,CISH was 64.81% （ 35/54 ） and 65.95% （ 31/47 ） . Amongst the cases of EGFR or TOPOⅡ protein overexpression (+++) group,the concordant ratios of IHC and CISH detecting EGFR or TOPOⅡ status were 80% and 83.33%; amongst the cases of EGFR or TOPOⅡ protein overexpression (++) group, the concordant ratios were 58.97% and 60%; amongst the cases of EGFR or TOPOⅡ protein expression (+/-) group, the concordant ratios were both 100%. The concordant ratios of HER2 detected by IHC, CISH was 100%． The gene status of HER2 detected by FISH. Though FISH detected HER2 gene Status in 107 cases of ESCC, gene amplifieation was found in 1 case,others were no gene amplifieation,incluing 1 case detected Chromosome 17 polysomy. Conclusion EGFR and TOPOⅡ are closely related to the occurrence and development of esophageal squamous cell carcinoma.their expressions all make a qualitative change in the esophageal high-grade intraepithelial neoplasia.
