乐复能对胰腺癌患者体内TNF-α的影响及其机制 李富军1邹益友1薛梅1 卢放根2（1中南大学湘雅医院消化内科，湖南长沙 410008；2中南大学湘雅二医院消化内科，湖南长沙 410011） 摘要：目的 观察乐复能对胰腺癌患者血清内TNF-α及其单核细胞内NF-κB mRNA表达的影响，以期探讨乐复能影响TNF-α分泌原因及其治疗胰腺癌的机制并为乐复能治疗其它免疫性疾病提供理论依据。方法 采用ELISA方法检测乐复能治疗前后胰腺癌患者血清内TNF-α的浓度，分离胰腺癌患者外周血单核细胞，采用RT-PCR方法检测单核细胞内NF-κB mRNA表达情况。结果 乐复能治疗前，胰腺癌患者血清内TNF-α的浓度为64.41±10.21 pg/ml，在给予乐复能治疗后，胰腺癌患者血清内TNF-α浓度为36.34± 0.92 pg/ml，下降约43.58%，治疗前后比较，P=0.00，差异有统计学意义。RT-PCR检测表明，乐复能治疗前后，胰腺癌患者外周血单核细胞内NF-κB表达明显下调（0.4994±0.0604 vs 0.2829±0.0365,p0.01）。结论 乐复能能明显抑制胰腺癌患者血清内TNF-α的分泌，下调其单核细胞内NF-κB的表达，其下调NF-κB表达作用可能与其抑制TNF-α分泌有关，而其抑制TNF-α分泌的作用可能与其治疗胰腺癌有关。 关键词： 乐复能；胰腺癌；肿瘤坏死因子；NF-κB Role and mechanism of novaferon on production of TNF-α in patients with pancreatic cancer LI Fu-jun1,XUE Mei1,LU Fang-gen2,ZOU Yi-you1(1Digestive Department of XiangYa Hospital Central South University, ChangSha Abstract:Aims To study the alteration of TNF-αin patients with pancreatic cancer before and after therapy with novaferon and the expression of NF-κB mRNA in monocytes in order to investigate the pathogenesis and treatment of pancreatic cancer and provide theoretical basis for immunity disease treatment.Methods To detect the concentration of TNF-αin patients with pancreatic cancer before and after therapy with novaferon by ELISA and the expression of NF-κB mRNA inmonocytes by RT-PCR.Results The concentration of TNF-αin patients was 64.41±10.21 pg/ml before treatment,and it decreased to 36.34± 0.92 pg/ml after treatment with novaferon，and the reduction rate was 43.58%，there was significant change with TNF-αin serum of patients before and after novaferon treatment(p=0.00) .there are also significant change about NF-κB mRNA expression in monocytes(0.4994±0.0604 vs 0.2829±0.0365,p0.01)Conclusion novaferon can downregulate TNF-αand the expression of NF-κB mRNA in patients with pancreatic cancer,and this fuction may be associated with pancreatic cancer treatment and TNF-αproduction.
