Clinical outcomes of papillary thyroid carcinoma vary. Most PTCs are associated with a good prognosis,however, in a certain group of patients it may have a fatal outcome. Therefor,it becomes more important to find reliable and consistent biomarkers for papillary thyroid carcinoma detection and monitoring.SPAG9 is suggested to have been involved in the regulation of migration, invasion,and metastasis of tumor cells.The usefulness of SPAG9 expression as a prognostic marker in papillary thyroid carcinoma has not been investigated yet. Objective To analyze the expression of SPAG9 in PTC as well as its association with the clinicopathologic features and prognostic significance. Methods SPAG9 protein expression was determined in papillary thyroid carcinoma cell lines in 79 papillary thyroid carcinoma specimens, 17 multinodular goiters (nonneoplastic hyperplasia), and 12 follicular adenoma tissue samples by immunohistochemistry. Results 1.The expression of SPAG9 was mainly located in the cytoplasm. Papillary thyroid carcinoma and follicular adenomas revealed inhomogeneous postive intensity while multinoduler goiters revealed weak SPAG9 protein. The positive expression rate of SPAG9 was100% in papillary thyroid carcinoma,which was significantly higher than that in benign thyroid lesions(P＜0.01). High expression of SPAG9 was detected in 65.82% of the cases.SPAG9 expression in papillary thyroid carcinoma was associated significantly with two clinicopathologic parameters,such as age (P=0.000),TNM stage (P=0.031).The expression of SPAG9 in papillary thyroid carcinoma had no signifiant correlation with gender , tumor size ,lymph node metastases ,tumor multicentricity and concurrent other thyroid diseases
