Abstract Although current research and development attention has been focused mainly on agricultural residues and dedicated energy crops such as corn stover and switchgrass, woody biomass (hard wood and soft wood) remains a very important feedstock for bioenergy production. This work focused on SPORL (Sulfite Pretreatment to Overcome Recalcitrance of Lignocellulose) and dilute-acid pretreatment for woody Biomass. The different effects of SPORL and dilute-acid pretreatment on earlywood and latewood cell walls of Douglas-fir were studyed firstly in this study. The results indicated that dilute acid pretreatment removed almost all the hemicelluloses, while SPORL pretreatment at initial pH 4.5 removed significant amount of lignin between 20~25%. But both are not sufficient for effective enzymatic saccharification for soft wood. SPORL at low initial pH combines the advantage of both dilute-acid and SPORL pretreatment at high pH to achieve approximately 90% hemicellulose removal and significant delignification of 10~20%. Results also showed that Wood density does not appear to contribute to wood recalcitrance and enzymatic saccharification. Cell wall chemical composition, especially lignin and hemicellulose content, contribute to cell wall recalcitrance. Forest residue from commercial plantation havesting and forest thinning operations is a substantial biomass resource that can be utilized for bioenergy production in China. While the quality of forest residue is low since the high bark content. This study investigated the bark effect on SPORL pretreatment and developed an effective strategy for collecting forest residues and converting to fermentable sugars. Pretreatment of Douglas-fir forest residues by SPORL and subsequent enzymatic hydrolysis revealed the presence of 14.3% bark can reduce substrate enzymatic digestibilities (SED) 16% compared with that from a bark free sample. The SED of a bark is 41% compared with 73% for wood when pretreated under the same conditions
