黄芪注射液对结肠癌SW480细胞生长抑制作用 　　[摘要] 目的：探索研究黄芪注射液对人结肠癌细胞株SW480细胞的生长抑制和凋亡作用机制。方法：实验设空白对照组、奥沙利铂对照组和黄芪注射液实验组（125、250、500、1 000 μg/ml）。MTT法检测黄芪注射液对SW480细胞生长增殖和凋亡的影响；流式细胞仪PI染色检测SW480细胞的凋亡。结果：与空白对照组相比，黄芪注射液对SW480细胞生长增殖有明显的抑制作用，且与作用浓度呈正比（P=0.0000.01）；流式细胞仪显示，黄芪注射液作用SW480细胞48 h后，125、250、500、1 000 μg/ml各组的凋亡率分别为24.2%、42.6%、64.1%、84.0%，较空白对照有明显升高，有显著性差异（P=0.0000.01）。结论：黄芪注射液能抑制SW480细胞生长，促进结肠癌细胞凋亡。 　　[关键词] 黄芪；SW480细胞；增殖；凋亡 　　[中图分类号] R73-3 [文献标识码] A [文章编号] 1673-7210（2011）06（c）-033-03 　　 　　The growth inhibitory effect of Astragalus Injection to the colon cancer cell line SW480 　　FAN Zhanbing, LI Ming, WEI Shuangjiang, QIN Caijuan, XIAO Qingwang 　　Department of General Surgery One, the People′s Hospital of Luanxian, Hebei Province, Luanxian 063700, China 　　[Abstract] Objective: To study the growth inhibitory effect and mechanism of Astragalus Injection to apoptosis of colon cancer SW480 cell lines． Methods: The control group, the oxaliplatin control group and the experimental group were included in this study. The influence of Astragalus Injection on the growth proliferation and apoptosis of SW480 cell in vitro was detected with MTT assay． The apoptosis was detected with flow cytometry (FCM) and PI staining． Results: The inhibition of the proliferation of SW480 cell lines in experimental group was more obviously compared with the control group, and which was directly proportional to the action concentration (P=0.0000.01)． FCM showed that the apoptosis rates in groups of 125 μg/ml, 250 μg/ml, 500 μg/ml, 1 000 μg/ml were 24.2%, 42.6%, 64.1%, 84.0% respectively after Astragalus Injection on SW480 cell after 48 hours.
