注射性rhBMP-22fPVP缓释液对糖尿病大鼠胫骨上段骨折愈合作用的实验研究.doc

只大鼠，分别进行右下肢 X 线片阅片观察评分和新生骨组织的 HE 染色、组织学 染色、用特异性方法测定血清中的 OC（ostepcalcin 骨钙素）表达、血清中的碱 性磷酸酶（alkaline osphatase，ALP）、及光镜下观察骨痂内 OB （osteoplast 成骨 细胞）数量，并描绘 BMP-2 释放曲线图。 结果： 骨折术后同一时间， X 线片检查结果示：各组局部的骨痂影均逐渐增加， 其中 A 组新生骨痂量均多于 B、C 组，而均 D 组少，D 组产生骨痂最多、愈合 最理想。生化检测指标显示：OC 浓度结果显示 D 组明显高于其他三组，ALP 浓 度结果显示 D 组 ALP 含量显著高于各组。随着骨修复的时间和程度的增大，局 部产生的 Ca 含量也相应的累积升高，含量统计结果显示 D＞A＞C＞B。组织学 切片结果观察显示： D 组骨小梁成熟度和密度均高于其余三组。免疫组化染色 后观察示：Ki67 指数（%）在术后的第 4 周达到峰值后成下降趋势，各时间点 数据显示：D 组峰值高于各组。苏木精-伊红染色光镜下观察显示，C 组 OB 数 量明显多于 B 组，略低于 A 组，明显低于 D 组。 结论： rhBMP-2 及 rhBMP-2/PVP 缓释液均能对糖尿病大鼠骨折处骨愈合起促进 作用。 PVP 溶剂对 rhBMP-2 有较理想的缓释作用， rhBMP-2/PVP 复合缓释液对 骨折愈合的促进作用优于局部 rhBMP-2 单纯作用。 关键词：骨形态发生蛋白2；聚乙烯吡咯啉酮；骨折愈合；糖尿病； 3 万方数据 Injection of rhBMP-2 / PVP slow-releasing composite to promote healing in tibial fracture on diabetic rats experiments Abstract Research objective: Through the establishment of diabetic rats in right tibia fracture model, to observe the local injections of rhBMP - 2 and injection of rhBMP - 2 / PVP bone repair material impact on diabetes than fracture healing in rats, and comparative research of liquid polyethylene pyrrole ketone solvents in diabetic rats in fracture healing slow-release effect of BMP.
