Methods After T cells enrichment, MACS separating system was used to sort CD8+CTL and CD4+CD25+Treg from spleen T lymphocytes. The optimal concentration of CTL and the best culture condition were explored by measuring the cytotoxicity of CTL agait MNK45 at different doses. Then CD4+CD25+ Treg were added into the culture system or in a separate well in trawell system. The influence of Treg on cytotoxicity of CTL was investigated by counting inactive MNK45 cells. Moreover, the cellular and immunological mechanism which might be involved was analyzed. Results In the culture system, CTL showed significant effect agait MNK45 (P). When ml (×104/ml) was added to this reaction system, the amount of killed MNK45 cells decrease obviously (P). And when Treg cells were cultured seperatly in the TraWell system, Treg cells must be increased ×105/ml to achieve the same inhibitory effect. Conclusio CD4+CD25+ T regulatory cells could exert inhibitory effect on CD8+CTL killing function in vitro, the celltocell contact mechanism plays important roles in mediating this effect. 【Key words】 Gastric tumor; Regulatory T cells; Cytotoxic T lymphocyte; MNK45 正常机体的抗肿瘤机制主要包括体液免疫和细胞免疫两方面，两者比较又以后者抗肿瘤活性为主导［1］。在抗肿瘤的细胞免疫机制中， 自然 杀伤细胞(natural killer cells，NK cells)和细胞毒性T淋巴细胞(cytotoxic T lymphocytes，CTL)作为抗肿瘤免疫的直接效应细胞，发挥着十分重要的作用。NK细胞和CD8+CTL在效应阶段均分别通过穿孔素、Fas/FasL、TNFα、LT(淋巴毒素)和INFγ等途径杀伤肿瘤靶细胞［2］。两类细胞的任何一类活性受到干预，都将影响机体的抗肿瘤效应。正常机体的调节性T细胞(regulat
