REV. A 1 OF 1 Flushing the 2475 Flow Cell When to Perform • Flush and passivate the flow cell after each shutdown, or when dirty. A dirty flow cell can cause baseline noise, decreased sample energy levels, calibration failure, and other problems with the operation of the 2475 Detector. Parts Required • None Tools Required • A wrench suitable for removing and replacing the column • A solvent miscible in both the mobile phase and water (methanol is commonly used) • HPLC-grade water • To passivate the flow cell, a strong cleaning solvent suitable for your system (6N nitric acid is commonly used) • A separate container for acid waste • A means for measuring the pH of acid effluent (if you use an acid as your cleaning solvent) NOTE: There is no “Flushing the 2475 Flow Cell” video. Flush and Passivate the Flow Cell 1. Stop mobile phase flow, then remove the column. 2. Replace the column with a union or section of tubing. 3. Attach tubing from the detector to the injector outlet where the column was removed. 4. Flush the mobile phase from the detector with a miscible solvent and water. 5. Flush the detector with HPLC-quality water to remove contaminants from the flow path. 6. Pump 6N nitric acid through the flow cell to clean the internal pathways. 7. Flush again with HPLC-quality water until the pH is neutral. 8. Reattach the column to resume flow of the mobile phase. Use an intermediary solvent if you are using a mobile phase that is not miscible in water. NOTE: Be certain that you do not exceed the pressure limit of the flow cell.
