响应面法优化(R)-α-羟基苯乙酸培养基#

摘要:目的: 采用响应面分析法对菌株 Bacillus sp.ULi-11 发酵产(R)-α-羟基苯乙酸培养
基进行优化。方法: 采用 Plackett-Burman 试验设计筛选出影响发酵产率的三个主要因素,采
用最陡爬坡路径逼近最大响应区域, 再用 Box-Behnken 试验设计及响应面分析法进行回归
分析。结果: 最佳培养基浓度分别为:酪蛋白胨 12.81g/L,淀粉 12.74g/L,KH2PO4 2.97g/L,
K2HPO4 1.2 g/L,MgSO4 0.15 g/L,CaCl2 0.125 g/L,MnSO4 0.015 g/L,ZnSO4 0.01 g/L。
在优化条件下,(R)-α-羟基苯乙酸产率达 69.78%,比优化前提高了 37.6%。结论: 响应
面分析法用于寻求(R)-α-羟基苯乙酸发酵的最佳培养基组分浓度是可行的。

Abstract: Purpose: Response Surface Methodology was applied to optimize the culture
components for (R)-α-hydroxyphenylacetic acid by Bacillus sp.ULi-11. Methods: By the initial
screening method, Plackett-Burman design was used to evaluate the influence of total eight related
factors. It showed that three factors playing the important roles in the medium. Path of steepest
ascent was undertaken to approach the optimal region of the fermentation conditions subsequently.
Finally, the concentrations of those three main factors were further optimized by using
Box-Behnken and Response Surface Analysis. Results: By solving the quadratic regression model
equation, the optimal concentrations of the variables were determined as: starch 12.81g/L, casein
peptone 12.74g/L, KH2PO4 2.97g/L, K2HPO4 1.2 g/L, MgSO4 0.15 g/L, CaCl2 0.125 g/L,
MnSO4 0.015 g/L, ZnSO4 0.01 g/L. The yield of (R)-α-hydroxyphenylacetic acid (69.78%) was
promoted by 37.6% at the optimal culture conditions, compared to that obtained from initial
fermentation conditions. Conclusion: It is available to find the optimal medium for
(R)-α-hydroxyphenylacetic acid by response surface methodology.
