Ltd ., Hohhot 011500, China) ABSTRACT: Objective To establish a rapid method for the determination of cholesterin in foods by RP-HPLC. Methods The samples were magnetically stirred and saponified in the water bath at 60 ℃ for 45 min, and extracted with petroleum ether and concentrated after making constant volume using anhydrous etha- nol. Then, the samples were separated on a DIONEX C 18 column (4.6 mm×250 mm, 5 μm) in elution with the mobile phase of methanol. Results The linearity of the method was 10∼400 μg/mL(r=1.000). The limit of detection was 20.6 μg/g with the relative standard deviations of 0.66%∼2.12%. The average recoveries were between 92.5%∼100.2%. Conclusion This method is rapid, accuracy, and precise, and it can meet the re- quirements of the cholesterol analysis.
