安定对大鼠局灶性脑梗死半影区神经细胞死亡的保护作用_医学论文 安定对大鼠局灶性脑梗死半影区神经细胞死亡的保护作用_医学论文 作者：吴中亮 刘飞 游思维 焦西英 鞠躬 【关键词】 脑缺血 关键词: 安定凋亡坏死脑缺血脑梗塞 摘 要:目的 研究安定对光化学诱发脑梗死后细胞死亡的保护作用. 方法 使用光化学诱发的大鼠局灶性脑缺血模型.采用原位末端标记技术（TUNEL）检测凋亡细胞.大鼠脑梗死术后存活3，6，12，24，48和72h，分别观察梗死灶坏死中心面积和凋亡细胞数目.术前24h开始腹腔注射安定（10mg#12539kg-1 #125398h-1 ），直到处死动物. 结果 TUNEL阳性细胞位于坏死中心与正常皮层之间的半影区.TUNEL阳性细胞的均数随着梗死后时间的延长逐渐增多，24h达到高峰.在24h时间点，安定治疗组梗死坏死中心的平均最大面积和TUNEL阳性细胞均数与对照组相比，明显减少（0.001）. 结论 安定可明显减少成年大鼠局灶性脑梗死后的细胞坏死和凋亡.该结果可为临床使用安定治疗缺血性脑血管病提供理论依据. Keywords:diazepamapoptosisnecrosiscerebral ischemiacerebral infarction 　　Abstract:AIM To study the protective effects of diazepam against nervous cell death after photochemically induced cere-bral ischemia.METHODS Focal cerebral ischemia induced by photothrombosis in adult rats was used as a model.Apop-tosis was detected by terminal deoxynucleotidyl transferase mediated dUTP-biotin nick end labeling（TUNEL）.The areas of necrotic cores and the numbers of apoptotic cells were studied quantitatively in the infarcted area at3，6，12，24，48and72h after operation，respectively.Repeated in-traperitoneal injections of diazepam（10mg#12539kg-1 #125398h-1 ）were given，starting at24h prior to the operation and contin-uing till the animals were sacrificed.RESULTS The TUNEL-positive cells were located in the transitional zone between the necrotic core and normal cortex.The mean num-ber of TUENL-positive cells was increased sharply when the postoperative time increased from3，6to12h and peaked at24h.The mean maximum area of the necrotic cores and mean number of TUNEL-positive cells at24h were signifi-cantly decreased（0.001）in the diazepam-treated experi-mental group than in the saline-treated control group.CONCLUSION Diazepam can significantly reduce both necrosis and apoptosis following cerebral ischemia induced by photothrombosis in adult rats.These findings provide a theo-retical warranty for the clinical use of diazepam in ischemic cerebrovascular diseases. 　　0 引言 　　近年来有研究显示，安定具有保护脑部缺血性神经元，防止其退行性变的作用［1，2］ .我们采用大
