玫瑰花聚丙烯酰胺凝胶电泳鉴别研究 李 明 山东省济南卫生学校，济南 250023 摘要 目的:建立不同栽培品种玫瑰花药材的聚丙烯酰胺凝胶电泳(PAGE)鉴别方法。 方法:蛋白质聚丙烯酰胺凝胶分析，电泳采用稳流控制，开始时电流15 mA，当样品到 达分离胶后，电流25 mA。以泳道中各谱带所在位置Pos为横坐标，谱带透光率OD为纵 坐标作图，绘制各谱带的透光率图并用 EDAS软件分析谱带相似度。结果：山东平阴的 “丰花”与 “重瓣”玫瑰花含有的蛋白质种类相似，含量相似；北京 “金顶”玫瑰花与 山东 “丰花”相似度较低；山东定陶的 “大红花”与 “丰花”相似度较低，蛋白质种类 有一定差异；阴干品与烘干品蛋白质电泳谱带透光位置有较大差异。结论:玫瑰花蛋白 质电泳的差异性表明了样品间蛋白质种类的差异，可作为玫瑰花栽培品种鉴别的依据； 加工温度对蛋白质影响很大。 关键词：玫瑰花；蛋白质-聚丙烯酰胺凝胶电泳；品种鉴别 * Identification of Flos Rosae Rugosas using polyacrylamide gel electrophoresis LI Ming Medical schools in Jinan, Shandong Province，Jinan 250023 Abstract Objective: To develop a new method for identification of different cultivated species of Flos Rosae Rugosas using polyacrylamide gel electrophoresis.Method: analytical method has been established using polyacrylamide gel electrophoresis(PAGE) . Electrophoresis , using steady flow control, starts with a beginning current of 15 mA, ends with a current of 25 mA when the sample reachs separation plastic . Laning in the location of the various bands position for abscissa, transmittance bands OD for mapping ordinate, we draw the map of bands ’light transmission rate and analyze banda similarity with EDAS software . Results:The results showed that the quantities and varieties of proteins in Fenghua Rosa which produced from Pingyin in Shandong province were similar to those of Chongban Rosa. Protein quantities and varieties in Beijing Jinding Rosa and Shandong Fenghua Rosa have a large difference. Protein varieties of Shandong Dahonghua Rosa and Fenghua Rosa were also different greatly .
