AbstractObjective o clarify the regulatory elements of Rad51 gene in its 5 -flanking region.
MethodsVarious constructs were obtained by cloning different DNA fragment s into pGL3 reporter
vector. hese construct s w ere then introduced into osteosarcoma cell line U2-OS by calcium phosphat e
method for transient expression of reporter gene and luciferase activit ies w ere measured by luciferase,
assay. ResultsCells transfected w ith pGL3 constructs containing fragment - 964 to + 1430 and - 733 to
+ 1430 show ed high luciferase activities. Obvious elevation of luciferase act ivities w as also observed in
3 - 964 - 412, - 746
cells transfected w it h pGL constructs containing four shorter derivative fragment s to
to - 412, - 651 to - 412 and - 536 to - 412. he highest luciferase activities w ere measured in
