62 2006;28(2) Effects of puer arin on L2type calcium channel in CA1pyramidal neurons in hippocampus of adult rats Tang Yu, Luo Rong2jing, Zhou Le2 quan ( Department of Physiology , The Basic Me ical College , GuangZhou Univer sity of TCM , GuangZhou 510405 ) Abstract Obj ective: To observe the effect of Puerarin on L2t ype calcium channel in CA1 pyrami al neurons in hippocampus of a2 ult rats aft er giving several concentrations. Methods: The pyrami al neurons in hippocampal CA1 region were issociate acutely from SD a ult rats by the enzymatical an automatical way. The cell suspension was then place into a 35mm petri ish . After allo2 wing the cell to settle , select ing the cell that accor with our experimental con itions. Then recor ing the ionic channel activity at the same hol ing voltage by the cel2l at tache of patch clamp techniques. Results: No significant effect were seen in L 2type calcium chan2 nel in CA 1 pyrami al neurons with the low concentrations of puerarin( 11 2, 21 4mmol# L- 1 ) . At a given hol ing voltage , significant - 1 change was observe in the open time of the L2t ype calcium channel after infusing Puerarin( 41 8, 91 6, 191 2mmol # L ) . Open time was 111 773 ? 41 838 ms for control , 71 948 ? 21 513ms for concentration of 41 8mmol L- 1 ( P 01 05, n= 5 ) , an 41 533 01 828ms ( 91 6mmol L- 1 , P 01 05 vs cont rol ) , then 31 042 ? 01 792ms( 191 2mmol # L- 1 , P 01 05 vs control ) . The open probability was 01 0011? 01 0001 in control , ecrease to 01 00053 ? 01 00018 ( 41 8mmol # L- 1 , P 01 05 vs cont rol ) , an 01 00032 ? 01 00013 ( 91 6mmol # L- 1 , P 01 05 vs cont rol ) , then went own 01 0002 1? 01 00009( 191 2mmol# L- 1 , P 01 05vs control ) . Conclution: Pu2 erarin can inhibit L - t ype calcium in CA 1 pyrami al neurons, ecrease the open ti
