药学学报 A cta Pharm aceu tica Sin ica 2008, 43 11 : 1071 - 1076 ·107 1 · ·综 　述 · 肠道 P糖蛋白辅料抑制剂的研究进展 闫　方 , 斯陆勤 , 黄建耿 , 李 　高 华中科技大学 同济医学院 药学院 , 湖北 武汉 430030 摘要 : 位于小肠上皮细胞顶端的 P糖蛋白 Pgp 是一种能量依赖性外排泵 ,可降低多种底物药物的生物利用 度 。抑制肠道 Pgp 以增加药物生物利用度受到越来越多的关注 。本文将肠道 Pgp 辅料抑制剂分为非离子型表面 β 活性剂 、聚乙二醇类 、 环糊精衍生物和壳聚糖硫醇盐 ,并对其作用机制进行综述 。与传统 Pgp 抑制剂相比,辅料抑 制剂的非特异性药理作用极小 ,因此几乎不产生副作用 。辅料抑制剂有望代替传统 Pgp 抑制剂 ,广泛用于增加难溶 性及低口服吸收药物的肠道吸收 ,从而增加该类药物的生物利用度 。这些辅料抑制 Pgp 的机制及其临床应用还需 更多研究和探讨 。 关键词 : P糖蛋白; 药用辅料 ; 抑制剂 中图分类号 : R943　　　文献标识码 : A 　　　文章编号 : 0513 - 4870 2008 11 - 1071 - 06 Advances in the study of exc ip ien t inh ib itors of in testina l Pglycoprote in YAN Fang, SI Luq in , HUAN G J iangeng, L I Gao S chool of P ha rm acy, Tongj iM ed ica l Colleg e, H uazhong Un ivers ity of S cience and Technology, W uhan 430030, Ch ina A b stract: Pglycop rotein Pgp located in the ap icalm emb rane s of in te stinal ab sorp tive cells is an energydep enden t efflux pump wh ich can reduce the b ioavailab ility of a w ide range of sub strate drugs. A cla ssification of excip ien t inh ib itors of in te stinal Pgp non ion ic su rfactan ts, po ly ethylene glyco l , β derivate s of cyclodextrin and th io lated ch ito san w ill be p re sen ted and then the inh ib ition m echan ism w ill be discu ssed. Comp ared w ith traditional Pgp inh ib itor, excip ien t inh ib itors app ear to have m in im al non sp ecific p harm aco logical activity, thu s poten tial side effects can be mo stly avo ided.
