Methods The rat carotid artery permanent occlusion (2VO) the establishment of VD animal model, SD rats were randomly divided into sham operation group, 2VO group and the puerarin group, each group was divided into 1 week, 3 weeks , 6 weeks, 2 months (n = 6) four time points; application of Y-water maze test of learning and memory ability of animals measured, using immunohistochemical staining of rat hippocampal CA1 area and the EPO protein expression of HIF1α . Results ① 2VO group and puerarin in rats Tour full time and strayed into blind-ended frequency were significantly increased, but at different time points of puerarin in rats learning and memory performance were significantly better than the 2VO group. ② each time point, 2VO group and the puerarin group and the hippocampal CA1 area HIF1α expression of EPO was significantly higher than those in sham-operated group, but the puerarin group, two kinds of protein expression was significantly lower than those in 2VO. Conclusion puerarin can increase within the ischemic brain tissue oxygen concentration, improving learning and memory function, VD with definite protective effect.
