Idiopathic thrombocytopenic purpura in children with the analysis of peripheral blood T-cell subtypes

PAGE \* MERGEFORMAT 19 Idiopathic thrombocytopenic purpura in children with the analysis of peripheral blood T-cell subtypes Authors: Xiao, Liu Fang, WU Chang-lin, Yang Meng Abstract T-cell subtype differentiation of drift may be related to the pathogenesis of some autoimmune disease mechanisms. This study was designed through analysis of idiopathic thrombocytopenic purpura (idiopathic thrombocytopenic purpura, ITP) in children with peripheral blood Th / Tc, Th1/Th2, Tc1/Tc2 cells equilibrium imbalance of T cell subsets in the pathogenesis of ITP in the role. Collected 30 cases of ITP in children with peripheral venous blood anticoagulant, isolation and purification were T cells to PE labeled anti-CRTH2 monoclonal antibody and Cy5 labeled anti-CD4, CD8 monoclonal antibodies for two-color flow cytometry, analysis ITP patients infants Th / Tc, Th1/Th2, Tc1/Tc2 percentage changes. The results showed that: ITP in children with peripheral blood T cells and compared to healthy children, Th cell percentage and Th / Tc ratio decreased significantly (P lt;0.05), Tc cells, no significant change in the percentage (Pgt; 0.05); Th1 and Th2 cells, the percentage of significantly decreased (P lt;0.05), Tc1 and Tc2 cells, no significant change in the percentage (Pgt; 0.05), Th1/Th2 and Tc1/Tc2 significantly higher proportion (P lt;0.05). Conclusion: ITP children that may exist in peripheral blood cell immune function and T cell subsets in low drift, Th1 and Tc1 cell ratio increased, showing clear advantages of Th1 type cells, this results suggest that abnormal differentiation of T smaller subsets play in the pathogenesis of ITP an important role.
