Methods The cultured human small-cell lung cancer cell line NCL  H446, MTT assay of nimesulide cytotoxicity; inverted microscope with Hoechst 33258 fluorescent staining of cell morphological changes; flow cytometry (FCM) analysis of apoptosis rate . MTT results showed Nimesulide dose-and time-dependent inhibition of human small cell lung cancer cell line NCL  H446 proliferation; inverted microscope shows cells smaller, become round, wrinkled, some cells detached floating in the medium. Visible under fluorescence microscopy dense chromatin condensed nuclei, fluorescence intensity increased, chromatin margination, nuclear fragmentation and apoptotic body formation. FCM results showed that nimesulide NCL  H446 can induce apoptosis, can also cause necrosis, 400μmol / L nimesulide for the role of cells, 24h after the early apoptosis rate was 7.65%, late apoptosis rate was 29.76%, accounting for 23.9% of necrosis. Conclusion nimesulide on cultured human small-cell lung cancer cell line NCL  H446 have more significant inhibition, the role and the induction of apoptosis and necrosis.
