 PAGE \* MERGEFORMAT 11 Oxaliplatin in human cervical cancer HeLa cell lines in vitro 【Abstract】 【Objective】 study of oxaliplatin on human cervical carcinoma HeLa cell line growth inhibition and on Caspase-3 activity. 【Methods】 employing HeLa cervical cancer cell lines by oxaliplatin treatment, MTT determination of cell growth inhibition rate, transmission electron microscopy observation of morphological changes, flow cytometry and in situ end labeling method (TUNEL) detection of cell apoptosis rate, colorimetric detection of Caspase-3 activity changes, and take advantage of AC-DEVD-CHO for Caspase-3 inhibition test. 【Results】 oxaliplatin (4 ~ 64 μmol / L) in vitro can be a time-and dose-dependent manner cervical cancer HeLa cells cytotoxicity and induced apoptosis, the role of cell line oxaliplatin after 48 h , the cells showed typical apoptotic morphological features; and to increase the HeLa cells, Caspase-3 activity, AC-DEVD-aldehyde (AC-DEVD-CHO) effectively inhibited oxaliplatin-induced apoptosis in HeLa cells. 【Conclusion】 oxaliplatin in vitro can inhibit the proliferation of human cervical cancer HeLa cells, and induce cell apoptosis, Caspase-3 activation is involved in oxaliplatin-induced apoptosis of HeLa cells.
