Methods: CCK-8 method Momordicin on K562/A02 the IC50 value of resistant cells, cell morphology and AnnexinV / PI double staining to detect apoptosis, flow cytometry determination of P-glycoprotein (P-glycoprotein, P-gp expression and caspase-3 and caspase-8 activity. Results: bitter resistance protein significantly inhibited cell K562/A02 proliferation, protein treated by bitter typical cell morphology change of apoptosis, AnnexinV / PI double staining showed significant increase in apoptotic cells, P-gp expression decreased, caspase-3 and caspase-8 activity was significantly increased. Conclusion: K562/A02 bitter resistance protein can induce apoptosis, reversal of drug resistance of leukemia cells with high expression of P-gp-mediated apoptosis suppression. Methods: The half-inhibition concentration (IC50 of momordin on K562/A02 cells was assessed by CCK-8 assay. Cell apoptosis was determined by Annexin V / PI double-staining and cytomorphology. P-glycoprotein (P-gp) expression, caspase-3 activity and caspase-8 activity were measured with flow cytometry (FCM). Results: Momordin obviously inhibited the proliferation of K562/A02 cells. A typical apoptotic change in the morphology was observed in K562/A02 cells after treatment with momordin. An obvious increase in the apoptosis rate of the cells was found with Annexin V / PI staining. And FCM showed P -gp expression obviously decreased while caspse-3 activity and caspase-8 activity markedly increased.
